Abstract
This invention concerns a covalent conjugate between a monosaccharide and/or glycan and the lipid carrier molecule phytol as well as a cell-free in vitro method and a cell free in vitro system for forming of a glycosylated peptide or polypeptide using said conjugate. A peptide or polypeptide comprising one or more glycosyl-moiety acceptor residues is contacted with a covalent conjugate between a monosaccharide and/or glycan and the lipid carrier molecule phytol in the presence of an oligosaccharyltransferase to form a glycosylated peptide or polypeptide. The invention also concerns a peptide or polypeptide with a glycosyl-moiety acceptor residue having an exogenous N-linked glycosylation sequence L(A/V)NYT, as well as an N-glycosylated conjugate of said peptide or polypeptide.
