Abstract

The invention relates to a whole blood compatible matrix for coupling a ligand and use in extracorporeal blood treatment, said matrix comprising a solid substrate, said substrate comprising a first layer of polyethylene glycol (PEG) and at least a second layer of polyethylene glycol, wherein said at least second layer is positioned outside the first layer and is bound to said first layer, and wherein the polyethylene glycol of the at least second layer comprises a terminal functional group for coupling said ligand to said polyethylene glycol. The invention further relates to a blood treatment device in the form of an adsorption cartridge or column, comprising the matrix according to the present invention, wherein the matrix is whole blood compatible. The invention further relates to the matrix of the invention comprising a ligand coupled thereto (apheresis matrix). The invention further relates to a method of preparing an apheresis matrix, comprising coupling the polyethylene glycol of the at least second layer of the matrix to the ligand. The invention further relates to a ligand coupled to the polyethylene glycol of the apheresis matrix according to the invention for use in the treatment of a medical condition by extracorporeal blood treatment.
