Abstract
The present invention relates to the treatment of fibrosis with a cell therapy based on myeloid cells. The invention provides myeloid cells, such as monocytes or macrophages that are resistant to M-CSF induced expression of a profibrotic gene signature. Said myeloid cells may be genetically modified in that the expression or activity of the transcription factors MAFB and/or MAF is reduced, inhibited or abolished. The administration of such myeloid cells to a subject leads to the reduction of fibrosis. Even more surprisingly, it is demonstrated by the invention that cell therapy with genetically engineered myeloid cells shows a dominant effect against the background of resident macrophages and monocytes after administration to mice in vivo. In particular, a dominant effect of Maf-DKO monocytes on the fibrosis-generating microenvironment of the lung is demonstrated, which indicates that these cells can also be used for the treatment of other fibrotic diseases.
