Abstract
The invention relates to the field of medicine, in particular to means and methods for the improvement, prevention or treatment of osteoarthritis. Provided is an oligonucleotide specifically hybridizing with miR-204 and a composition for ameliorating, preventing, or treating osteoarthritis, including the same. The oligonucleotide includes 20 to 22 linked nucleotides and consists of a first region, a second region, and a third region in order from a 5' end to a 3' end, wherein the first region includes three consecutively connected nucleotides, each complementary to a tail region of miR-204 and including a constrained sugar modification, the second region includes three connected nucleotides connected, the nucleotides included in the second region each independently being nucleotides including a constrained sugar modification or a non-constrained 2'-substituted sugar modification, and the third region includes a plurality of connected nucleotides, the nucleotides included in the third region each independently being nucleotides including a non-constrained 2'-substituted sugar modification, and wherein the oligonucleotide includes 30% or less of nucleotides including a constrained sugar modification based on a total number of nucleotides.
