Abstract

The present invention relates to a composition for predicting the responsiveness to cetuximab in cancer, more specifically, a composition comprising a complex, wherein the complex comprises: a peptide represented by the amino acid sequence of SEQ ID NO: 1, which is cleaved by a lysosomal enzyme present in a cancer cell; a fluorophore and a quencher bound to the peptide; and cetuximab bound to the C-terminus of the peptide. The Cetux-probe of the present invention operates via the same mechanism of action as cetuximab, enabling rapid and accurate prediction of cetuximab's therapeutic efficacy and responsiveness. The activated fluorescence of the Cetux-probe can be used to measure EGFR degradation, and a strong linear correlation with cetuximab's cytotoxicity was observed in colorectal cancer cells and in mice bearing tumors. Furthermore, it was confirmed that the predictive capability of the activated fluorescence of the Cetux-probe is significantly higher than that of EGFR expression or KRAS mutation status. Therefore, the Cetux-probe of the present invention can be effectively utilized to predict responsiveness to cetuximab therapy by evaluating EGFR degradation.
