Abstract

The present invention provides a cell having improved histocompatibility with a transplant recipient. Specifically, by knocking in a polynucleotide encoding a single-chain fusion peptide mimicking HLA-G and/or HLA-E that functions in an inhibitory manner against the activity (cytotoxicity) of NK cells or macrophages into a region encoding an alpha chain of an HLA Class I molecule, the expression of the alpha chain of the HLA Class I molecule is suppressed while the single-chain fusion peptide is expressed.
