Abstract

Short synthetic peptides derived separately from interleukin-4 (IL-4) and stromal cell-derived factor-1 (SDF-1/CXCL12), designed by structure-guided selection of receptor-binding regions (IL-4(5-15), IL-4(78-92), SDF-1(1-8), SDF-1(39-49)). When applied to mammalian cells at approximately 250 ng/mL, these peptides reproduce selected biological activities of the parent cytokines without causing off-target differentiation. IL-4-derived peptides enhance myoblast fusion and reduce adipogenic differentiation; SDF-1-derived peptides promote migration of fibroblasts, myoblasts, mesenchymal stromal cells and induced pluripotent stem cells. The peptides are non-toxic and suitable as components of cell culture media or biomaterial scaffolds for skeletal muscle regeneration.
