Abstract

A ligation method including: ligating, to a 3' end of each of plural kinds of nucleic acids having different sequences, a 5'-adenylated single-stranded DNA that is a 3' adapter in which a base sequence of a 5' end excluding an adenylic acid at the 5' end is any one of base sequences shown in Table A; and after the ligating of the 5'-adenylated single-stranded DNA, ligating, to a 5' end of each of the plural kinds of nucleic acids, a single-stranded nucleic acid that is a 5' adapter; a method of suppressing adapter dimer formation; a method of preparing an NGS library; an NGS library preparation kit; and a 3' adapter.
