Abstract
The present disclosure provides a universal, high-throughput antibody discovery platform based on: (i) a negative-selection method for enriching B cells or plasma B cells without relying on plasma B cell surface biomarkers, and (ii) a microfluidic droplet-based functional screening system capable of identifying single cells secreting antigen-specific antibodies. The enrichment method employs a customizable cocktail of antibodies that target non-B lineage immune cells, followed by magnetic bead depletion. Enriched antibody-secreting cells are subsequently encapsulated into picoliter or nanoliter droplets together with (a) antigen-coated beads, (b) a fluorescent labeled antigen with immunoglobulin-capture antibody-coated beads, or (c) antigen-expressing reporter cells, allowing for functional detection of secreted antibodies inside each droplet. Through multi-channel fluorescence detection-incorporating detection antibody signals, dead-cell exclusion dyes, and optional blocking-antibody controls-droplets containing antigen-specific antibody-secreting cells are isolated. The recovered cells undergo single-cell BCR amplification or bulk BCR sequencing to obtain naturally paired heavy and light chain sequences.
